Effects of Shallot Extract and Sprout Extract on the In Vitro Growth of Stevia (Stevia rebaudiana B.) Using MS Medium and a Simplified Medium

This study investigated the effects of red onion extract and mung bean sprout extract on the in vitro growth of Stevia (Stevia rebaudiana B.). Stevia is a low-calorie natural sweetener with great potential, but conventional propagation methods are often inefficient. Tissue culture offers a solution, although standard MS media can be costly. Therefore, this research also explored the use of a simpler, low-cost alternative medium. The study involved three experimental trials. Results showed that red onion extract produced the highest number of shoots—up to 11—surpassing both synthetic PGR Benzyl Amino Purine (BAP) and sprout extract. Activated charcoal helped reduce browning, and the simple medium made of agar, Gandasil B, and water showed better sterility than MS-based media. While mung bean sprout extract had some positive effects, results were inconsistent. Explant length and node number also affected shoot growth. Overall, red onion extract with a simple medium was the effective treatment.
