The Effect of Wholeblood Storage with K₃EDTA and Na₂EDTA Anticoagulants on Leukocyte Count, Neutrophil Count, and Leukocyte Morphology

Abstract

Hematological examinations play a crucial role in disease diagnosis and are influenced by pre-analytical factors, such as the type of anticoagulant and storage temperature. EDTA is widely used due to its minimal impact on blood cell morphology. This study aims to determine the effect of whole blood storage using K₃EDTA vacutainer and conventional Na₂EDTA anticoagulants on leukocyte count, neutrophil count, and leukocyte morphology. This was a laboratory-based cross sectional experimental study involving D4 TLM students at UMSIDA as respondents. Samples were stored at room temperature and cold temperature, and examined at three hours after blood collection. Cell counts were analyzed using a Hematology Analyzer,while morphology was observed through peripheral blood smears. The results no significant differences in leukocyte and neutrophil counts based on the type of anticoagulant, storage temperature, or storage duration (p>0.05).Morphological changes began to appear after 12 hours at room temperature and after 24 hours under cold storage.
