The detection of Toxoplasma gondii in cat using the Microscopic and Loop-Mediated Isothermal Amplification (LAMP) method at the Larangan Market Sidoarjo

Toxoplasmosis, caused by Toxoplasma gondii, is a zoonotic disease affecting both humans and animals, particularly those with weakened immune systems. This study aimed to detect T. gondii in cat feces using saturated NaCl microscopy and the Loop-Mediated Isothermal Amplification (LAMP) method, while also evaluating the sensitivity of LAMP. Samples were collected using accidental sampling over one month in August. A descriptive analysis compared microscopy and LAMP results using the SPSS version 23 kappa test. Of the 9 samples from Larangan Market, Sidoarjo, 4 tested positive and 5 negative. Detection involved NaCl flotation, DNA isolation via the resin method, and LAMP with visual color indicators. Sensitivity testing used fivefold PBS dilution to improve DNA quality. The results showed perfect agreement between methods (k = 1.000, p < 0.05), indicating high reliability. Further studies should increase sample size and diversity to strengthen population representation and result validity.
