Abstract

Lead (Pb) is a toxic heavy metal that accumulates and causes problems. Moringa (Moringa oleifera Lamk.) seeds came from Sedayulawas Village, Lamongan Regency. This study assessed Moringa seed extract effectiveness in reducing blood Pb levels in Wistar rats and determining the dose. Conducted in May–June 2026 at Universitas Muhammadiyah Sidoarjo, the study used a pre-test post-test control group design. Twenty-four rats were divided into six groups: negative control, positive control, 0.5% Na-CMC control, and extract groups receiving 1000, 1250, or 1500 mg/kg BW. Except for the negative control, rats received lead acetate (700 mg/kg BW) for seven days, followed by 14 days of treatment. Blood Pb was measured by AAS and analyzed using the Wilcoxon test in SPSS 25. The extract contained alkaloids, flavonoids, saponins, steroids, triterpenoids, phenolics, and tannins. Results showed no significant overall effect (p=0.068; p>0.05). Reduction occurred at 1500 mg/kg BW, but the optimal dose remained undetermined.
