Analysis of the Use of the NS1 Antigen Detection Method and Loop-Mediated Isothermal Amplification (LAMP) in the Diagnosis of Dengue Hemorrhagic Fever (DHF)

Abstract

Dengue Hemorrhagic Fever (DHF) is an infectious disease caused by the dengue virus (DENV) which is transmitted through the bite of Aedes aegypti and Aedes albopictus mosquitoes. Early diagnosis can be done using the NS1 and LAMP methods. This study aims to analyze the use of the NS1 antigen detection method and RT-LAMP in the diagnosis of DHF. The study used a descriptive exploratory design on 30 blood samples of suspected DHF patients. The examination was carried out using an immunochromatography-based NS1 antigen RDT and RT-LAMP with RNA extraction using the spin column method. The RT-LAMP examination in this study showed a higher number of positive samples than the NS1 method. The Cohen's Kappa test showed a p value = 0.039, which indicates low agreement between the two methods. RT-LAMP has a higher detection capacity so it has the potential to be an early diagnosis method for DHF.
