Ascariasis is a disease caused by a soil-transmitted helminth infection specifically, the parasite Ascaris lumbricoides that affects health, particularly in preschool-aged children with compromised immune systems. This study aimed to detect A. lumbricoides in the stool samples of preschoolers using the formalin-ether sedimentation microscopy method and the Loop-Mediated Isothermal Amplification (LAMP). Samples were collected using the total sampling technique over a two-week period in April. Of the 30 stool samples analyzed, the microscopic method identified 24 positive and 6 negative samples for A. lumbricoides; some of these samples were co-infected or triple-infected with Strongyloides, Schistosoma, Trichuris trichiura, Toxoplasma, and Taenia sp. The LAMP method identified 26 positive and 4 negative samples. In a concordance test using SPSS with the Kappa test, the results showed near perfect agreement with a value of (k=0.762) and (p<0.05).
