Abstract

Leukocyte Differential Count (DLC) is influenced by pre-analytical factors, including anticoagulant type and sample storage duration. This study aimed to evaluate the effect of K₃EDTA and Na₂EDTA on DLC results using automated and manual methods after storage at 2–8°C. A laboratory experimental study with a repeated-measures design was conducted using whole blood samples from six healthy male subjects aged 17–25 years. Samples were analyzed at 0, 24, and 60 hours using a hematology analyzer and peripheral blood smear microscopy. Statistical analysis showed no significant differences (p > 0.05) in lymphocyte, MID cell, and granulocyte values based on storage duration, anticoagulant type, or analytical method. Although mild leukocyte morphological changes were observed after prolonged storage, these changes did not significantly affect DLC results. Therefore, whole blood samples anticoagulated with K₃EDTA or Na₂EDTA remained stable for Differential Leukocyte Count examination after storage at 2–8°C for up to 60 hours.
