Comparison of the Effectiveness of K3EDTA and Na2EDTA Anticoagulants on the Stability of Differential Leukocyte Count (DLC) Results Using Manual and Automated Methods Whole Blood Storage

The differential leukocyte count is a hematological test used to determine the percentage of leukocyte types, aiding in clinical diagnosis. Result accuracy is influenced by pre-analytical factors. This study aimed to analyze the effect of storage duration on whole blood samples using K3EDTA and Na2EDTA anticoagulants on differential leukocyte count results obtained via manual and automated methods. This laboratory-based experimental study employed a cross-sectional design using venous blood samples from six respondents, analyzed via peripheral blood smears and a hematology analyzer at various storage intervals. Two-way analysis revealed no significant effect (p>0.05) on lymphocyte and granulocyte parameters. Kruskal–Wallis test results indicated that storage duration and testing method significantly influenced mid-cell parameters (p<0.05), whereas the type of anticoagulant did not (p>0.05). It is concluded that storage duration and testing method affect differential leukocyte count results, while the use of K3EDTA versus Na2EDTA anticoagulants does not yield a significant difference.
