The Effect of Whole Blood Storage Time with K₃EDTA and Na₂EDTA Anticoagulants on Differential Leukocyte Count Results Using Manual and Automatic Methods

Abstract

Differential leukocyte count (DLC) is a hematological examination affected by preanalytical factors, including storage time and anticoagulant type. This study evaluated the effect of storing whole blood with K₃EDTA and Na₂EDTA on DLC results using manual and automated methods. A cross-sectional laboratory experiment was conducted in April 2026 involving six D4 Medical Laboratory Technology students at Muhammadiyah University of Sidoarjo. Blood samples were analyzed immediately (0 hours) and after 36 and 72 hours of refrigerated storage (2–8°C) using Na₂EDTA and K₃EDTA anticoagulants. DLC parameters included lymphocytes, MID-Cells, and granulocytes. Storage time and anticoagulant type showed no significant effect on all DLC parameters (p > 0.05), whereas the examination method significantly affected MID-Cells (p < 0.05). These findings indicate that both Na₂EDTA and K₃EDTA maintain DLC stability for up to 72 hours under refrigerated conditions, although leukocyte morphological alterations may occur during prolonged storage.
