Thalassemia is an inherited blood disorder caused defect hemoglobin synthesis hemolysis disruption globin chain formation, leading to hemolytic anemia and changes in erythrocyte morphology. This study aims determine the correlation between MCV, MCH, MCHC, and EHDT values patients thalassemia. The study design quantitative analysis using a cross-sectional approach involving 50 patient with thalassemia at Dr. Saiful Anwar Regional General Hospital in East Java Province, in June 2026. The results of the Spearman correlation test showed a very strong positive correlation between MCV and MCH (r=0.891,p=0.000), positive correlation between MCH and MCHC (r=0.347,p=0.014), no significant correlation between MCV and MCHC (r=0.113,p= 0.433). Decreased erythrocyte indices correlated with EHDT erythrocyte morphological of microcytic hypochromic anemia, of target cells, teardrop cells, elliptocytes, and ovalocytes. Concluded a decrease in erythrocyte indices is positively correlated with and reflects a failure in globin chain synthesis, as evidenced by abnormal erythrocyte morphology identified through EHDT examination.
